The EZBlue™ Cell Assay Kit is based on reduction of the water soluble, redox dye EZBlue™. The dye itself is dark blue in color and has intrinsic fluorescence. The fluorescence changes when EZBlue™ is reduced by metabolically active cells to the pink resorufin product. The spectral properties of resorufin allows the molecule to be detected using either absorbance or fluorescence.

EZkillTM Mycoplasma Elimination Kit is an antibiotic based elimination kit designed to eliminate mycoplasma from cell culture. Mycoplasma contamination of cell culture is a major problem in both research and industrial production. Ideally it is recommended to discard the contaminated culture after autoclaving. Only in cases where cell line is very important and non-replaceable, treatment for elimination should be considered. Antibiotic based method is the most common and effective method with no marked cytotoxicity.

The EZdetectTM DAPI stain kit has been designed for detection of mycoplasma based on rapid uptake of the fluorescent stain DAPI by cellular DNA. DAPI selectively binds minor grooves of the DNA. The excitation maximum of dsDNA-bound DAPI is 358nm and emission maximum is 461nm. Mycoplasma staining with DAPI appears as a fine particulate or filamentous staining over the cytoplasm at 100X magnification. Nuclei of the cells are also brightly stained by this method and thereby act as endogenous positive control for the staining procedure.

It selectively binds minor grooves of the DNA and hence allows for efficient staining of the nuclei. Hoechst stain can be excited either with a xenon mercury arc lamp or a UV laser and is detected through a blue filter. Mycoplasma staining with Hoechst appears as a fine particulate or filamentous staining over the cytoplasm at 100X magnification. Nuclei of the cells are also brightly stained by this method and thereby act as endogenous positive control for the staining procedure.

This kit has been designed for detection of mycoplasma based on amplification of spacer region between 16S and 23S rRNA genomic DNA sequence. This sequence is highly conserved between various mycoplasma species. This allows for detection of commonly encountered cell culture contaminants. Eukaryotic DNA is not amplified. A successful PCR reaction is indicated by a distinct band of 350-400bp derived from the internal control DNA on the agarose gel.

Melondialdehyde(MDA) is one of such TBARS used for estimation of lipid peroxidation. This assay is based on the reaction of MDA with a chromogenic agent thiobarbituric acid(TBA) at high temperature and acidic conditions to form MDA-TBA adduct(1:2). This complex has absorption maximum at 532nm and can be measured spectrophotometrically between 530 - 540nm. Absorbance is directly proportional to the TBARS concentration in the sample.

EZAssayTM GST Activity Estimation kit is simple and convenient tool for sensitive and reproducible detection and quantitation of GST activity in an array of samples like serum, cell lysates and tissue homogenates. This assay is based on the GST-catalyzed reaction between reduced glutathione and the GST substrate 1-chloro-2,4-dinitrobeneze(CDNB). CDNB can react with the broadest range of GST isoenzymes. The conjugation of glutathione to the CDNB substrate results in an increase in absorbance at 340nm.

The EZcountTM BrdU Cell Assay kit is designed for determination of cell proliferation and / or effect ofcytotoxic agent. When cells are cultured with labeling medium containing Pyrimidine analogue 5-bromo-2’-deoxyuridine(BrdU), BrdU gets incorporated in the DNA of proliferating cells in place of thymidine. BrdU labelled proliferating cells can be detected using Anti-BrdU antibody. The labeling medium is removed, and cells are fixed and DNA is denatured using fixation/denaturation solution to make BrdU accessible to the Anti-BrdU-HRP antibody. HRP substrate TMB is added for color development. The magnitude of the absorbance for the developed color directly correlates to the quantity of BrdU incorporated into cells, which is a direct indication of cell proliferation.
