IL4 is a pleiotropic cytokine produced by activated T cells. IL4 is a ligand for interleukin 4 receptor. The interleukin 4 receptor also binds to IL13, which may contribute to many overlapping functions of this cytokine and IL13. STAT6, a signal transducer and activator of transcription, has been shown to play a central role in mediating the immune regulatory signal of this cytokine.
This gene, IL3, IL5, IL13, and CSF2 form a cytokine gene cluster on chromosome 5q, with this gene particularly close to IL13. IL4, IL13 and IL5 are found to be regulated coordinately by several long-range regulatory elements in an over 120 kilobase range on the chromosome. Two alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported.

IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells. IL-13 is involved in several stages of B-cell maturation and differentiation. It up-regulates CD23 and MHC class II expression, and promotes IgE isotype switching of B cells. This cytokine down-regulates macrophage activity, thereby inhibits the production of pro-inflammatory cytokines and chemokines. This cytokine is found to be critical to the pathogenesis of allergen-induced asthma but operates through mechanisms independent of IgE and eosinophils.

IL12 stimulates the production of IFNgamma from T-cells and natural killer cells, and also induces differentiation of Th1 helper cells. Interleukin 2 (IL-2) is a powerful immune-regulatory lymphokine produced by T-cells in response to antigenic or mitogenic stimulation. IL-2/IL-2R signaling is required for T-cell proliferation and other fundamental functions which are essential for the immune response. IL-2 stimulates growth and differentiation of B-cells, NK-cells, lymphokine activated killer cells, monocytes, macrophages, and oligodendrocytes. Interleukin 15 is a protein that regulates T and natural killer cell activation and proliferation. As a pleiotropic cytokine, it plays an important role in innate and adaptive immunity. Cytokines and interleukines share many biological activities.
IL-15 is constitutively expressed by a large number of tissues and cell types, including monocytes, macrophages, dendritic cells (DC), keratinocytes, fibroblasts and nerve cells.
They are found to bind common hematopoietin receptor subunits, and may compete for the same receptor, and thus negatively regulate each other's activity. The number of CD8+ memory cells is shown to be controlled by a balance between this cytokine and IL2. This cytokine induces the activation of JAK kinases, as well as the phosphorylation and activation of transcription activators STAT3, STAT5, and STAT6.
Studies of the mouse counterpart suggested that this cytokine may increase the expression of apoptosis inhibitor BCL2L1/BCL-x(L), possibly through the transcription activation activity of STAT6, and thus prevent apoptosis. Two alternatively spliced transcript variants of this gene encoding the same protein have been reported.

IL10 is a cytokine produced primarily by monocytes and to a lesser extent by TH2-lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation.
IL 10 inhibits the synthesis of a number of cytokines, including IFN-gamma, IL-2, IL-3, TNF and GM-CSF produced by activated macrophages and by helper T-cells. It inhibits the formation of cytokines responsible for the Th1 answer thereby it promotes the survival, increase and the antibody production of B-lymphocytes.
It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway.

IL-3 is a potent growth promoting cytokine that promotes the survival, differentiation and proliferation of a broad range of hematopoetic cell types.
It is produced by T cells, mast cells, and eosinophils and enhances thrombopoieses, phagocytosis, and antibody-mediated cellular cytotoxicity. Because of its ability to activate monocytes, it is supposed to have additional immunoregulatory roles. Many of the activities mentioned above are dependent on co-stimulation with other cytokines.
